To do business is a euphemism used in the Continental, meaning to murder another person; usually a target in a hitjob. Doing business on Continental grounds is against the rules, and results in the offender having their privileges revoked and ultimately executed.

In John Wick: Chapter 2, after having a $7 million hit placed on him, John Wick kills Santino D'Antonio at the Continental Hotel, breaking the rule. Rather than having him executed on the spot, Winston gives Wick a marker and a one-hour grace period before making him excommunicado.
